Chemically induced pluripotent stem cells (CiPSCs): a transgene-free approach.
Induced pluripotent stem cells (iPSCs) could be generated by a single gene Oct4 and chemical compounds, in which exogenous expression of Oct4 was indispensable for reprogramming. Recent advances in chemical-mediated cellular reprogramming suggest that small molecules alone (i.e. without Yamanaka factors) can successfully establish iPSCs from mouse somatic cells.

متن کاملDerivation of Transgene‐Free Rat Induced Pluripotent Stem Cells Approximating the Quality of Embryonic Stem Cells
Although a variety of reprogramming strategies have been reported to create transgene-free induced pluripotent stem (iPS) cells from differentiated cell sources, a fundamental question still remains: Can we generate safe iPS cells that have the full spectrum of features of corresponding embryonic stem (ES) cells? Studies in transgene-free mouse iPS cells have indicated a positive answer to this...
متن کاملInduced Pluripotent Stem Cells: Challenges and Opportunities
Regenerative capacity of mammals is limited and can rarely regenerate a specific organ or tissue fully. Due to these limitations, regenerative medicine seeks efficient and safe cell sources for regeneration of damaged tissues and organs or treatment for incurable diseases. Human embryonic stem cells (HESCs) hold two important properties called self renewal and pluripotency. However, the use of ...
متن کاملNo factor left behind: generation of transgene-free induced pluripotent stem cells.
Induced pluripotent stem cells (iPSCs) hold great potential for regenerative medicine, yet their implementation in the clinic still seems far out of reach due to concerns about their safety. The development of safe and efficient reprogramming methods is a critical step towards clinical application of iPSCs. Recently, much progress has been made in the technology of generating iPSCs that are fre...
